If you just want to mock something up, you can use a pre-built UI system / component framework and just put together UIs declaratively, without having to worry about the underlying CSS or HTML at all. Flask is simple and lightweight and as you said it give you flexibility. But if you want to have something that give you more flexibility and control over everything else besides the routing and server loop I would suggest bottle.
