There's a great plugin for After Effects called Duik Bassel 2, it allows to rig and animate characters easily, it's made for motion graphics but for your comic you could also export your characters as images after you pose them. Source: almost 2 years ago

Zbrush is the gold standard and it's not cheap. It has recently switched to subscription only. Zbrush also offers a stripped down free version for beginners and dabblers called Zbrush Core Mini.
