There's a great plugin for After Effects called Duik Bassel 2, it allows to rig and animate characters easily, it's made for motion graphics but for your comic you could also export your characters as images after you pose them. Source: over 1 year ago

I think Spine is more optimized for video games, you can achieve a very similar look with more flexibility with something like After Effects with the Duik plugin, or maybe even Live2D. Source: about 3 years ago
