Download the free and excellent GrandPerspective app and scan your disk, it will show you in a nested grahical form what is taking space. Maybe it's a sleep image, maybe it's on-disk TM backups cos you haven't connected your TM disk in a while, maybe it's some crazy big cache... GP will let you know so you can fix it.
